Looking at ASME QW-253 and QW-253.1 your existing SMAW WPS for P1 to P8 using a F5 covered electrode will serve your stated purpose. No requalification of your WPS is necessary.
Your WPS allows you to use P1 material with F5 covered electrode for welder qualification. This will qualify them for P1 and P8 material, ref QW-423.1
Your customer may want a WPS that clearly specifies the material being welded, you example 316L(P8) to A285(P1). In this case you just develop a new WPS for your customer from your existing PQR.

I need to qualify welders to weld P43(Ni-Cr-FE tube) to a P4 (ASTM A217 WC6) using SFA 5.11 E8N12 (ENiCrFe-3, F43)
Looking at QW-353 (Base metals QW-403.18) I can substitute QW-423 in its place?, and use A P1 plate for testing?
and QW-404.15 Filler metals I need to use an F43 filler? (or use QW-433 and use F34 or any F41 thru F45 filler).

You are correct. You can qualify for Nickel Alloy SMAW using F43 Filler and P-1 base material.

Was the original PQR, the one with A240 to A36, done with approved ASME materials. Meaning the specification of the material. Were they SA-36(P1) and SA-240 Type......? (P8)? If they were ASTM A-36(S-1), and A-240 Type.........?(S-8), then you might want to look at QW-420.2 S-Numbers.
The third paragraph states: If the original PQR materials were assigned S-numbers or S-number Group number, then the range quailified is limited to S-number and S-number Group numbers. (i.e., qualification using P-Number material qualifies corrosponding S-number materials; qualification using an S-number material quailifies corrosponding S-number materials but, not corrosponding P-number materials)
