In 50mm thk plate, what will be the depth of groove joint, smaw process, we want implement.

Basically, it's up to you.
There are good reasons to alter bevel depth, depending on external circumstances such as design load, access, distortion, weld process, etc...
Questions are:

Are you working to a particular code or standard?
Does the joint require complete weld penetration?
Do you intend to back gouge?
What type of material?

Some codes have pre-qualified procedures that specify joint dimensions.
Most have a process detailed for qualifying reliable procedures based on proposed joint conditions.

I am working in pressure vessel, sec viii div 1 code.
Job is intended for back gauging.
Material is carbon steel.

Ok, then it's pretty simple...
Get your most experienced welders and fitters together and ask them to provide recommendations on joint bevel dimensions that will be the most efficient while helping to ensure the final weld will pass RT.
Get your Engineer to verify that the final recommendation is compliant with the vessel design loads.
Then qualify the weld procedure in accordance with the specifications detailed in ASME Section IX.
If the procedure test plate qualifies, write a WPS that details exactly how to replicate the qualified weld, and train your welders in its use.
Get your AI to review and approve or reject the procedure documents before starting fabrication.
If the procedure test plate fails, start over.
