I'm an amateur welder with a 250 Amp stick machine. I need to make a grill for a barbeque pit but have never welded stainless before.

Any tips or suggestions for which sticks would be best? Any special preparations necessary? What about types of stainless that would be best suited? I'm thinking 1/4 or 3/8 round bar.

Yes just get you some 309 stainless rod and That will work just fine with your stick mach.

To avoid rusting, you should use type 316 material and E316, 3/32 stainless steel electrodes.
