I have inspection for vessel fabrication as follow BS PD 5500 , We would like to clarify that the nozzle to main shell need to perform MPI or DPI before close reinforcing pad or not.Or we can do after completed weld at reinforcing pad.

If you are using AWS welding symbols per AWS A2.4, you can use multiple reference lines to indicate the nozzle to shell weld, then NDT, then the reinforcement weld, and then the final NDT. The other method would be to include notes in the tail of the welding symbol directing the reader to the applicable notes on the drawing.

Your problem is not clear to me but it sounds like you want to perform MPI on nozzle attachment to assure complete penetration. If this is the case then one side should be welded, then the back side gouged, MPI'd, then continue welding. To put the reinforcement plate on before or later will depend on where you start your welding from.
